Detailed description

Ubash3a-m3 mice contain a ZFN-mediated mutation targeted to exon 9 of ubiquitin associated and SH3 domain containing, A (Ubash3a) gene, creating a 7 bp deletion and early termination. UBASH3A belongs to the T-cell ubiquitin ligand (TULA) family along with UBASH3B. UBASH3A expression is restricted to lymphoid tissues, specifically T cells, where it is capable of suppressing T cell receptor signaling. UBASH3A deficiency accelerates insulitis and type 1 diabetes (T1D) progression in both sexes by 10 weeks of age.

Development

This zinc finger nuclease induced mutation targets exon 9 of the ubiquitin associated and SH3 domain containing, A (Ubash3a) gene, creating a 7 bp deletion and early termination. The mRNA encoding the ZFN was injected into NOD/ShiLtJ one cell embryos and one mouse was identified and bred back to a NOD/ShiLtJ mice. Resulting Ubash3a-m3 offspring were bred to NOD/ShiLtJ mice. Upon arrival at The Jackson Laboratory, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize NOD/ShiLtJ oocytes (Stock No. 001976).
