Rosa26^{fl-Stop-fl-Rab7} mice express full-length mouse Rab7 (RAB7, member RAS oncogene family) cDNA upon cre recombinase-mediated excision of a loxP-flanked Stop cassette. When bred with Cd19-cre mice (Stock No. 006785) to generate mice with B cell-specific overexpression of RAB7, antibody responses are increased, concomitant with increased activation-induced cytidine deaminase (AID) expression and class switch DNA recombination (CSR). Homozygotes do not appear to be viable.

Detailed description

Rab7 (RAB7, member RAS oncogene family) is an integral component of the B cell NF-κβ activation machinery involved with immunoglobulin class switching, as central to the maturation of antibody and autoantibody responses.

Rosa26^{fl-Stop-fl-Rab7} mice express full-length mouse Rab7 cDNA upon cre recombinase-mediated excision of a loxP-flanked Stop cassette. When bred with Cd19-cre mice (Stock No. 006785) to generate mice with B cell-specific overexpression of RAB7, antibody responses are increased, concomitant with increased activation-induced cytidine deaminase (AID) expression and class switch DNA recombination (CSR).

To date, no homozygotes have been recovered from heterozygote x heterozygote crosses.

Development

A loxP-Stop-FRT-reverse neo-FRT-loxP-CMV promoter-Rab7 cDNA cassette was introduced to intron 1 of the mouse Gt(ROSA)26Sor gene via homologous recombination in C57BL/6-derived embryonic stem (ES) cells. The Rab7 cDNA is full-length and derived from mouse. Resultant chimeric mice were crossed with a C57BL/6 background strain expressing Flp recombinase to excise the neomycin cassette, leaving the Stop cassette flanked by loxP sites. This strain was maintained on a C57BL/6 genetic background by the donating laboratory (see SNP note below).

In 2021, a 48 SNP (single nucleotide polymorphism) panel analysis, with 43 markers covering all 19 chromosomes and the X chromosome, as well as 5 markers that distinguish between the C57BL/6J and C57BL/6N substrains, was performed on the rederived living colony at The Jackson Laboratory Repository. Two of the 5 markers that determine C57BL/6J from C57BL/6N were found to be segregating, including the rd8 recessive mutation associated with retinal degeneration on Chromosome 1 within the Crb1 locus. These data suggest the mice sent to The Jackson Laboratory Repository were on a mixed C57BL/6N;C57BL/6J genetic background.
