Description

Nolz1^fl/fl mice have loxP sites flanking exon 3 of the zinc finger protein 503 (Zfp503) gene. These mice may be useful when studying dopaminergic axon guidance and innervation.

Detailed description

Nolz1^fl/fl mice have loxP sites flanking exon 3 of the zinc finger protein 503 (Zfp503) gene. Also known as Nolz1, this gene encodes a transcriptional regulator expressed in the ventral tegmental area dopaminergic (DA) neuronal lineage and striatal projection neurons. Mice that are homozygous for this floxed allele are viable and fertile. When bred to mice that express tissue-specific Cre recombinase, resulting offspring will have exon 3 deleted in the cre-expressing tissues. For example, when bred to mice expressing Cre recombinase in the forebrain, resulting Nolz1^-/- mice have DA axons that are misguided and fail to innervate the striatum.

Development

The Nolz1^fl targeting vector was designed to insert loxP sites flanking exon 3 of the zinc finger protein 503 (Zfp503) gene. An FRT-flanked neomycin resistance (neo) cassette was also inserted between exons 2 and 3. The construct was electroporated into C57BL/6-derived embryonic stem (ES) cells and correctly targeted ES cells were injected into recipient blastocysts. The resulting male chimeric mice were bred to FLP-expressing mice on a C57BL/6 background to remove the neo cassette. Progeny were crossed to remove the FLP-expressing transgene and resulting Nolz1^fl/fl mice were then backcrossed to C57BL/6J mice (Stock No. 000664) for at least 6 generations by the donating lab. Cryopreserved sperm was sent to The Jackson Laboratory. Upon arrival an aliquot was used to fertilize C57BL/6J oocytes to establish our live colony.
