CXCL8 (chemokine (C-X-C motif) ligand 8) encodes a proinflammatory cytokine also known as IL8 (interleukin 8). IL8 is secreted by multiple cells including mononuclear macrophages, neutrophils, T lymphocytes, epithelial cells and fibroblasts. IL8 is involved in the systemic inflammatory response syndrome and is upregulated in patients with discogenic back pain. pCALL-hIL8 transgenic mice conditionally express human CXCL8 under the control of the widespread CAG promoter. The transgene includes a lacZ-neomycin construct flanked by loxP sites inserted upstream of CXCL8 cDNA. The donating investigator confirms that, in the absence of cre,&nbsp; betagalactosidase is widely expressed in these mice. When bred to mice that express tissue-specific Cre recombinase, resulting offspring will have lacZ-neo deleted in cre-expressing tissues allowing expression of human CXCL8. Hemizygous pCALL-hIL8 mice are viable and fertile. Homozygous mice appear small and weak, fertility has not been assessed. For example, when bred to mice with Cre recombinase expression in synovial joints and intervertebral disc tissue (GDF5-Cre) offspring exhibit lower nesting behavior, increased immobility, decreased eating, and reduced travel and speed.
