Description

Pitx1/Cre transgenic mice express cre recombinase under the control of the mouse Pitx1 (paired-like homeodomain transcription factor 1) promoter. Cre is expressed in the craniofacial and oral ectoderm as early as E9.5 before the formation of Rathke's pouch, so it is useful for targeting the nascent anterior pituitary gland before the specification of hormone producing cell types.

Detailed description

Pitx1/Cre transgenic mice express cre recombinase under the control of the mouse Pitx1 (paired-like homeodomain transcription factor 1) promoter. Cre is expressed in the craniofacial and oral ectoderm as early as E9.5 before the formation of Rathke's pouch, so it is useful for targeting the nascent anterior pituitary gland before the specification of hormone producing cell types.

Development

The -8 kb promoter of the mouse Pitx1 gene drives expression of cre recombinase in this transgenic strain. The mutation was created in CB6F1 (BALB/c x C57BL/6) embryos. Resultant mice were backcrossed to C57BL/6NCrl mice for 9 generations by the donating laboratory (see SNP note below). The transgenic integration site and copy number are unknown (May, 2020).

In 2020, a 48 SNP (single nucleotide polymorphism) panel analysis, with 43 markers covering all 19 chromosomes and the X chromosome, as well as 5 markers that distinguish between the C57BL/6J and C57BL/6N substrains, was performed on the males sent to The Jackson Laboratory Repository. One of the 43 markers, on Chromosome 3, was found to be segregating with an unknown source. Also, 1 of the 5 markers that determine C57BL/6J from C57BL/6N were found to be segregating. These data suggest the mice sent to The Jackson Laboratory Repository were on a mixed C57BL/6N;C57BL/6J genetic background.
