Description

Peak1^flox mutant mice possess loxP sites flanking exon 4 of the Peak1 (pseudopodium-enriched atypical kinase 1) gene and may be useful in generating conditional mutations to study the VEGFR2 axis, blood vessel formation, and cancer.

Detailed description

Peak1 (pseudopodium-enriched atypical kinase 1) encodes a widely expressed non-receptor tyrosine kinase that is involved in the regulation of angiogenesis through the PEAK1-GATA2-VEGFR2 signaling axis. PEAK1 is dysregulated in many cancers. These conditional Peak1^flox mice possess loxP sites flanking exon 4 of the Peak1 gene. Mice that are homozygous for this floxed allele are viable and fertile. When bred to mice that express tissue-specific Cre recombinase, resulting offspring will have the exon 4 deleted in cre-expressing tissues.

For example, when bred to mice with Cre recombinase expression in early mouse embryos (Tg(EIIa-cre)C5379Lmgd, Stock No. 003724), homozygous knockout offspring exhibit a reduction of the retinal superficial vessel plexus and reduced branch points in the retinal vasculature at P7. Examination of aortic tissue from KO adult mice reveals a reduction in de novo aortic vessel formation and reduced number and length of aortic sprout length.

 

Development

The targeting construct was designed to insert a loxP site upstream of exon 4 and a loxP-flanked neomycin cassette downstream of exon 4. The construct was electroporated into C57BL/6NTac-derived iTL IC1 embryonic stem (ES) cells. Correctly targeted ES cells were injected into blastocysts. Chimeric males were crossed to C57BL/6J females. Offspring, were bred with mice carrying an unspecified Flp recombinase to delete the neo cassette. The resulting progeny were crossed to remove the Flp-expressing transgene. The colony was crossed to C57BL/6J for approximately 10 generations. Upon arrival, mice were bred to C57BL/6J (000664) for at least one generation to establish the colony.
