Description

Exons 6 and 7 of the mouse Glp1r (glucagon-like peptide 1 receptor; also known as GLP-1R) gene are flanked by loxP sites in these GLP-1r conditional mutant mice. Cre recombinase-mediated excision of the floxed exons results in a knock-out allele, useful in studies of metabolism.

Detailed description

Glp1r (glucagon-like peptide 1 receptor; also known as GLP-1R) encodes a peptide hormone that is released from the gut in response to nutrient ingestion and that has a range of metabolic effects, including enhancing insulin secretion and decreasing food intake.

Exons 6 and 7 of the mouse Glp1r gene are flanked by loxP sites in these GLP-1r conditional mutant mice. Cre recombinase-mediated excision of the floxed exons results in a knock-out allele.

Development

An FRT-flanked neomycin resistance cassette with a 5' loxP site was inserted upstream of exon 6 and an additional loxP site was inserted downstream of exon 7 via homologous recombination in C57BL6/J-derived embryonic stem (ES) cells. Crosses with B6;SJL-Tg(ACTFLPe)9205Dym/J mice (Stock No. 003800) were carried out to excise the neomycin resistance cassette, leaving exons 6 and 7 flanked by loxP sites. Resultant mice were backcrossed to C57BL/6J for 5-6 generations by the donating laboratory (see SNP note below).

Upon arrival at The Jackson Laboratory, sperm was cryopreserved. Cryorecovery/live colony development will involve C57BL/6J (Stock No. 000664).

In 2023, a 48 SNP (single nucleotide polymorphism) panel analysis, with 43 markers covering all 19 chromosomes and the X chromosome, as well as 5 markers that distinguish between the C57BL/6J and C57BL/6N substrains, was performed on the mice sent to The Jackson Laboratory Repository. One of the 43 markers throughout the genome was segregating with an unknown source on Chromosome 2. Also, 2 of the 5 markers that determine C57BL/6J from C57BL/6N were found to be segregating. These data suggest the mice sent to The Jackson Laboratory Repository were on a mixed C57BL/6 genetic background and/or incompletely backcrossed.
