Detailed description

MLR10 transgenic mice exhibit cre recombinase expression in both the lens epithelium and lens fibers of the eye under the transcriptional control of the mouse Cryaa (crystallin, alpha A) promoter modified by the insertion of a Pax6 consensus binding element/lens epithelial cell enhancer. Cre expression is not found in other parts of the eye.

Development

The approximately 3 kb CPV14/Cre transgenic construct, composed of cre recombinase under the transcriptional control of the mouse Cryaa promoter modified with a Pax6 consensus binding element/lens epithelial cell enhancer (at position -86 of the promoter), was injected into FVB/N embryos. This line was maintained on an FVB/N genetic background by the donating laboratory (see SNP note below). This is founder line MLR10, which demonstrates expression in both the lens epithelium and lens fiber cells, but not in other parts of the eye.

In 2021, a 48 SNP (single nucleotide polymorphism) panel analysis, with 43 markers covering all 19 chromosomes and the X chromosome, as well as 5 markers that distinguish between the C57BL/6J and C57BL/6N substrains, was performed on the males sent to The Jackson Laboratory Repository. Six of the 43 markers throughout the genome were segregating with an unknown source. These data suggest the mice sent to The Jackson Laboratory Repository were on a mixed genetic background. Albino and agouti coat color alleles may be anticipated.

Molecular note: The expression of sequence encoding cre recombinase was driven by a modified alpha A crystallin promoter. The promoter consists of -282/+43 of the mouse Cryaa gene with an inserted copy of Pax6 consensus binding site in a 5' to 3' orientation at position -86 of the promoter. Seven lines were created with this line having expression of the transgene in both the lens epithelium and lens fiber cells but not in other parts of the eye.
