Human myeloperoxidase (hMPO) is considered a myeloid-specific protein, present at high levels in circulating neutrophils and monocytes. Recent findings show that the MPO gene can be atypically/aberrantly expressed in some non-myeloid cells under stress conditions such as astrocytes and neurons in Alzheimer's disease and astrocytes in a 1-methyl-4-phenyl-1,2,3,6-tetrahydropyrine (MPTP)-induced model of Parkinson's disease, as well as some cancer cells. A functional hMPO promoter polymorphism, -463G, increases hMPO expression and has been linked to a risk for Alzheimer's disease, cardiovascular disease, lung cancer, and some epithelial cancers (PMID: <a href="https://www.ncbi.nlm.nih.gov/pubmed/?term=31175983">31175983</a>). The polymorphism has also been associated with an incidence or severity of inflammatory diseases, including atherosclerosis and Alzheimer's disease, and some cancers. hMPO is aberrantly expressed in macrophages (atherosclerosis), breast and ovarian cancer, intestinal epithelial cells (colitis), MPO-ANCA (antineutrophil cytoplasmic antibodies), multiple sclerosis, and neurons, astrocytes, and microglia (Alzheimer's and Parkinson's disease). The polymorphism is within an Alu element encoding four hexamer repeats recognized by nuclear receptors (AluRRE). The mouse Mpo gene lacks the primate-specific AluRRE. These MPOG (hMPO) transgenic mice express the intact human myeloperoxidase gene bearing the -463G (G allele) promotor polymorphism, along with introns and flanking regions within a single 32 kilo base insert. The -463G polymorphism creates an Aci1 restriction site not found in the -463A polymorphism. A single integration of the transgene is located on the mouse X Chromosome. The hMPO transgene is expressed as in humans, with high levels in myeloid cell precursors and some monocyte/macrophages.&nbsp; When hMPO mice are crossed to an APP (amyloid beta (A4) precursor protein ) transgenic model of Alzheimer's Disease (APP23, <a href="http://www.informatics.jax.org/allele/MGI:2447146">Tg(Thy1-APP)3Somm</a>, as in Stock No. <a href="https://www.jax.org/strain/030504">030504</a>), the hMPO transgene is atypically expressed robustly in astrocytes. When combined with the Tg(THY1-SNCA*A53T)M53Sud (Stock No. <a href="https://www.jax.org/strain/008135">008135</a>) mutation, mice exhibit worsened motor impairment, and an earlier onset of paralysis.
