Description

HA-RARalpha KI knock-in mice express hemagglutinin-tagged mouse RARA (retinoic acid receptor, alpha) protein. Cre recombinase-mediated excision of floxed exons 4 and 5 results in the production of hemagglutinin-tagged RARA protein lacking the DNA binding domain.

Detailed description

HA-RARalpha KI knock-in mice express hemagglutinin-tagged mouse RARA (retinoic acid receptor, alpha) protein. Cre recombinase-mediated excision of floxed exons 4 and 5 results in the production of hemagglutinin-tagged RARA protein lacking the DNA binding domain. Heterozygotes and homozygotes are viable and without overt phenotype. Homozygotes are not fertile. The HA tag can be detected by immunoblot.

Development

A loxP site was introduced to intron 3, an FRT-neo-FRT-loxP cassette was placed in intron 5, and two hemagglutinin (HA) tags were inserted, in-frame, into exon 6 via homologous recombination in C57BL/6J x 129S6 F1 hybrid embryonic stem cells. The FRT-flanked neomycin cassette was excised through crosses with mice expressing Flp recombinase, leaving exons 4 and 5 (encoding DNA binding domains B and C) flanked by loxP sites in addition to the flag-tagged exon 6. The donating laboratory reported that the resultant mice were backcrossed to C57BL/6J for more than six generations (see SNP results below) prior to sending 2 males (agouti coat color) to The Jackson Laboratory Repository in 2022.

In 2022, a 48 SNP (single nucleotide polymorphism) panel analysis, with 43 markers covering all 19 chromosomes and the X chromosome, as well as 5 markers that distinguish between the C57BL/6J and C57BL/6N substrains, was performed on the 2 males sent to The Jackson Laboratory Repository. A mix of C57BL/6 and 129 substrains in addition to an unknown strain contribution on Chromosome 13 were detected. These data suggest the mice sent to The Jackson Laboratory Repository were on a mixed STOCK genetic background.
