Detailed description

CARMA3^F/F mice possess loxP sites flanking exons 1-3 of the caspase recruitment domain family, member 10 (Card10) gene. Card10 encodes CARMA3, a member of the membrane-associated guanylate kinase (MAGUK) family that has been found to be expressed in in the heart, liver, kidney, and lung, specifically in airway epithelial cells (AECs). CARMA3 activates the signaling of the transcription factor, NF-κB, which is involved in inflammatory cell development and responses. In the lung, NF-κB is involved in aspects of asthma pathogenesis, including cytokine and mucin production from epithelial cells, epithelial cell barrier function, and airway remodeling. CARMA3 has also been reported to be over-expressed in various cancer types that include breast, glioma, colon and non-small-cell lung cancers.

Mice that are homozygous for this floxed allele are viable and fertile. When bred to mice that express tissue-specific Cre recombinase, resulting offspring will have exons 1-3 deleted in the Cre-expressing tissues.

When bred to mice expressing cre recombinase throughout the tracheal epithelium, the bronchiolar epithelium, and in a subset of distal alveolar cells, resulting SPC^Cre/CARMA3^F/F mice exhibit reduced airway eosinophilia and proinflammatory cytokine production, impaired dendritic cell maturation in the lung, and impaired Ag processing in dendritic cells.

Development

The CARMA3^F targeting vector was designed to insert a frt-flanked neomycin selection cassette, followed by a loxP site, upstream of exon 1 and a second loxP site, downstream of exon 3 of the caspase recruitment domain family, member 10 (Card10) gene. The construct was electroporated into (C57BL/6NTac x 129S6/SvEvTac)F1-derived iTL BA1 embryonic stem (ES) cells and correctly targeted ES cells were injected into recipient blastocysts from C57BL/6 females. The resulting chimeric mice were bred with actin-flp expressing mice to remove the neo cassette, and resulting mice were subsequently bred together to remove the FLP expressing transgene. Offspring were further bred to C57BL/6NJ mice for a total of at least 9 generations. Upon arrival at The Jackson Laboratory, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6NJ oocytes (Stock No. 005304).
