Description

The entire coding region of the mouse Pou4f2 gene was replaced with a neomycin cassette in this Brn-3.2^- knock-out allele. Adult mice demonstrate a loss of approximately 60-70% of retinal ganglion cells.

Detailed description

Pou4f2 (POU domain, class 4, transcription factor 2; also called Brn-3.2) critically modulates the terminal differentiation of distinct sensorineural cells.

The entire coding region of the mouse Pou4f2 gene was replaced with a neomycin cassette in this Brn-3.2^- knock-out allele. Immunohistochemical analysis on sections of newborn dorsal root ganglia of homozygous mice confirmed that no detectable protein was expressed from this allele.

Heterozygotes and homozygotes do not display any obvious behavioral defects, develop to adulthood, and are fertile. Although the overall structure of the eye is preserved in homozygotes, mature (P17-P120) retinal whole mounts show a loss of approximately 60-70% of retinal ganglion cells. Axonal pathfinding is defective and retinal ganglion neurons exhibit survival defects. Neonate profiles are normal.

Development

A neomycin resistance cassette replaces a 2.7 kb genomic fragment containing the entire coding sequence of the gene. The mutation was created via homologous recombination in 129S4/SvJae-derived J1-derived embryonic stem (ES) cells. Resultant mice were backcrossed to C57BL/6NCrl for more than 10 generations by the donating laboratory.

In 2020, a 48 SNP (single nucleotide polymorphism) panel analysis, with 43 markers covering all 19 chromosomes and the X chromosome, as well as 5 markers that distinguish between the C57BL/6J and C57BL/6N substrains, was performed on the males sent to The Jackson Laboratory Repository. Four of the 5 markers that determine C57BL/6J from C57BL/6N were found to be segregating, on Chromosomes 6, 8, 15, and the rd8 recessive mutation associated with retinal degeneration on Chromosome 1 within the Crb1 locus. These data suggest the mice sent to The Jackson Laboratory Repository were on a mixed C57BL/6N;C57BL/6J genetic background.
