Description

Clec5a knock-out mice contain a neomycim cassette designed to replace exon 2 of the Clec5a (C-type lectin domain family 5, member a) gene. Exposure to long term cigarette smoke in homozygous mice results in reduced alveolar macrophage activation and pulmonary pathologies. This strain may be useful for studying chronic obstructive pulmonary disease (COPD), host-pathogen interactions, and macrophage functions.

Detailed description

Clec5a (C-type lectin domain family 5, member a) encodes a type II transmembrane protein that interacts with the adaptor protein TYROBP (TYRO protein tyrosine kinase binding protein or DAP12) and plays a role in cell activation. Clec5a is implicated in the progression of multiple acute and chronic inflammatory diseases. Clec5a knock-out (KO) mice contain a neomycin cassette designed to replace exon 2.  Mice that are homozygous for the targeted mutation are viable and fertile. Clec5a^-/-  mice exposed to long term (6 month) cigarette smoke exhibit a decrease in LPS-induced alveolar macrophage responsiveness, decreased airspace enlargement, decreased lung inflammation, decreased levels of TNFA and IL6, and fewer macrophages (and less activated macrophages). These mice may be useful for studying chronic obstructive pulmonary disease, host-pathogen interactions, and macrophage functions.

Development

A targeting vector containing a neomycin resistance cassette was used to replace exon 2. The construct was electroporated into 129-derived embryonic stem (ES) cells. Correctly targeted ES cells were injected into blastocysts. The resulting chimeric animals were crossed to C57BL/6J females, offspring were backcrossed to C57BL/6J for at least 10 generations. Upon arrival, mice were bred to C57BL/6J for at least one generation to establish the colony.
