Human ATP6AP2 (ATPase H+ transporting accessory protein 2; also called PRR, (pro)renin receptor) is a component of the renin-angiotensin system that mediates angiotensin II (ANG II) formation in the central nervous system (CNS). Local renin-angiotensin systems (RASs) are present within various tissues and have important physiological and pathophysiological roles in cardiovascular regulation. PRR overexpression mediates both ANG II-dependent and -independent reactive oxygen species (ROS) formation and NADPH oxidase (NOX) activation in cultured murine neurons.Syn-hPRR transgenic mice express full-length human ATP6AP2 (ATPase H+ transporting accessory protein 2; also called hPRR) under the control of the rat neuron-specific synapsin 1 (Syn1) promoter. Line 57994-1 exhibits high levels of hPRR mRNA in the brain, but not in other tissues, including the heart, kidneys, lung, liver, spleen, muscles, blood vessels, white adipose tissue and pancreas.Human prorenin cannot cleave mouse angiotensinogen, consequently, administration of human prorenin to Syn-hPRR mice cannot activate the ANG II-dependent pathway and instead selectively activates ANGII-independent PRR signaling in neurons. Overexpression of neuronal human (pro)renin receptor mediates angiotensin II-independent blood pressure regulation in the central nervous system. This creates a novel model for investigating the importance of these pathways in the neural control of blood pressure in vivo. Intracerebroventricular infusion of human prorenin induces an elevation of blood pressure in Syn-hPRR mice that is independent of ANG II formation, but required for phosphorylation of ERK1/2 and activation of NADPH oxidase 4 (NOX4) in the central nervous system.
