Description

IL-22-eGFP mice express an enhanced green fluorescent protein (EGFP) under the control of the Il22 (interleukin 22) promoter. IL22 is produced by group 3 innate lymphoid cells (ILC3s) in the intestine and CD4+ T cells and is associated with chronic intestinal inflammation or infection.  This mutant mouse strain may be useful in visualizing IL22 production by ILC3s and its relationship to inflammatory bowel disease.

Detailed description

The IL-22-eGFP transgene uses the mouse Il22 (interleukin 22) regulatory elements to associate expression of the enhanced green fluorescent protein (EGFP) with IL22, in particular, IL22 production by group 3 innate lymphoid cells (ILC3s) and CD4+ T cells in the gastrointestinal tract. In the intestine, IL22 is involved in host-microbe interactions and is associated with inflammatory bowel disease, sepsis, and cancer. EGFP expression reflects IL22 protein production by ILC3s. Mice hemizygous for the transgene are viable and fertile, homozygote viability/fertility has not been assessed. This mutant mouse strain may be useful in visualizing IL22 production by ILC3s and other immune cells.

Development

The bacterial artificial chromosome, RP24-227B3, containing the entire Il22 gene plus upstream (30 kb) and downstream (2 kb) regulatory sequences was modified to insert EGFP cDNA downstream of the Il22 exon 1 followed by an FRT-flanked neomycin cassette. The neomycin cassette was later removed using FLP recombinase leaving one FRT site. The BAC was microinjected into the pronuclei of fertilized C57BL/6J oocytes. Founder line 1 was bred to C57BL/6J mice. These IL-22-eGFP mice were subsequently maintained by breeding transgenic mice with C57BL/6J. Upon arrival, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes (Stock No. 000664).

In 2020, a 48 SNP (single nucleotide polymorphism) panel analysis, with 43 markers covering all 19 chromosomes and the X chromosome, as well as 5 markers that distinguish between the C57BL/6J and C57BL/6N substrains, was performed on the males sent to The Jackson Laboratory Repository. Two of the 5 markers that determine C57BL/6J from C57BL/6N were found to be segregating. These data suggest the mice sent to The Jackson Laboratory Repository were on a mixed C57BL/6N;C57BL/6J genetic background.
