Description

Nur77^RFP BAC transgenic mice express turbo red/orange fluorescent protein (tRFP) from the mouse Nr4a1 (nuclear receptor subfamily 4, group A, member 1; Nur77) promoter. These mice are useful in studying thymocyte selection as well as lymphocyte activation, and can be used as a reporter of T cell receptor signaling strength.

Detailed description

Nur77^RFP BAC transgenic mice express turbo red/orange fluorescent protein (tRFP) from the mouse Nr4a1 (nuclear receptor subfamily 4, group A, member 1; Nur77) promoter. tRFP, derived from sea anemone Entacmaea quadricolor, demonstrates fast maturation and is known for its high photostability, pH stability, and bright fluorescence (excitation/emission maxima are 553 and 574 nm, respectively). These mice are useful in studying thymocyte selection as well as lymphocyte activation, and can be used as a reporter of T cell receptor signaling strength.

Nur77^RFP hemizygotes are viable, fertile and without overt phenotype. The viability and fertility of homozygotes has not been assessed (February, 2020).

The Nur77^RFP (red fluorescent) BAC transgenic vector used to make this strain was adapted from the vector used to create Nur77^GFP (green fluorescent) Stock Nos. 018974 and 016617.

Development

The Nur77^RFP (red fluorescent) BAC transgenic vector used to make this strain was adapted from the vector used to create Nur77^GFP (green fluorescent) Stock Nos. 018974 and 016617.

In brief, the original Nur77^GFP transgenic vector was made by introducing a Cre recombinase/green fluorescent protein (GFP) fusion cassette to the 5'end of the full-length Nr4a1 (Nur77) gene encoded by bacterial artificial chromosome (BAC) clone RP24-366J14.

The new Nur77^RFP vector was introduced to C57BL/6J embryos. Resultant mice were backcrossed to both C57BL/6J and C57BL/6N by the donating laboratory. The insertion site and copy number of the transgene are not known. Maintenance of this strain at The Jackson Laboratory will likely involve C57BL/6J (Stock No. 000664).
