Dhps (deoxyhypusine synthase) has been studied largely in the context of cancer, where it promotes oncogenesis by enabling the production of proteins involved in cellular migration and growth. The gene has also been found to impact the proliferation of &beta; cells in the pancreas, influencing glucose homeostasis. Conventional knock-outs of the mouse Dhps gene result in embryonic lethality.Exons 2-7 of the mouse Dhps gene are flanked by loxP sites in these Dhpslox mice. Cre recombinase-mediated excision of the floxed region results in a knock-out allele.Tamoxifen-induced progeny derived from crosses with MIP1-CreERT (Tg(Ins1-cre/ERT)1Lphi; Stock No. <a href="https://www.jax.org/strain/024709" target="_blank">024709</a>) mice develop a pancreatic &beta; cell-specific knock-out of Dhps. High fat diet-fed knock-out mice develop diabetes with significant glucose intolerance and hyperglycemia compared to high fat diet-fed controls. &beta; cell proliferation is impaired.
