Description

KIF9 CKO mutant mice possess loxP sites flanking exon 3 of the Kif9 (kinesin family member 9) gene and may be useful in generating conditional mutations to study male fertility.

Detailed description

KIF9 (kinesin family member 9) encodes a protein involved in the regulation of the degradation of the extracellular matrix, and in lower eukaryotes KIF9 homologs play a role in ciliary motility. Kif9 mRNA expression is observed in epithelia containing multi-ciliated cells such as ependyma, bronchioles and oviduct, but is highest in testis. These conditional KIF9 CKO mice possess loxP sites flanking exon 3 of the Kif9 gene. Mice that are homozygous for the mutation are viable and fertile. When bred to mice that express tissue-specific Cre recombinase, resulting offspring will have the exon 3 deleted in cre-expressing tissues. These mice may be useful for studying male fertility.

For example, when bred to mice with Cre recombinase expression in the female germline (C57BL/6-Tg(Zp3-cre)92Knw/J), male homozygous knockout offspring develop sperm with defective flagellar motility and are sterile.

Development

CRISPR/Cas9 endonuclease-mediated genome editing was used to insert loxP sites in intron 2 and intron 3 (creating a floxed exon 3). A pair of guide RNA and Cas9 endonuclease mRNA were introduced into C57BL/6J-derived fertilized eggs by pronuclear injection along with a donor plasmid carrying around 2 kB homologous arms on each side of the floxed exon 3 and a primer sequence for genotyping. Embryos were transferred to pseudopregnant females, and correctly targeted pups were bred to C57BL/6J mice for germline transmission. Founders carrying the loxP sites were identified by PCR and sequencing. Upon arrival, mice were bred to C57BL/6J (Stock No. 000664) for at least 1 generation to establish the colony.
