Twist1 (twist basic helix-loop-helix transcription factor 1) encodes a transcription factor that is involved in embryogenesis and is overexpressed in human cancers. Overexpression of Twist1 is associated with tumor cell invasion and metastasis. Twist1-tetO7-luc mice carry a bidirectional Tet-responsive promoter directing expression of Twist1 and luciferase. Transgene expression can be assessed by measuring luciferase using bioluminescence imaging. Mice homozygous for the transgene are viable and fertile. When mated to a mutant strain expressing tetracycline-controlled transactivator protein (tTA or rtTA), expression of TWIST1 protein may be regulated with tetracycline or its analog doxycycline (dox) in the double mutant offspring. These transgenic mice may be useful for research in tumor metastasis. When bred to mice carrying Tg(Cebpb-tTA)5Bjd (Stock No. <a href="https://www.jax.org/strain/003563"">003563</a>) and Tg(tetO-MYC)36Bop (Stock No. <a href="https://www.jax.org/strain/034532"">034532</a>) dox-induced progeny expressing Myc and Twist1 in the liver develop hepatocellular carcinoma (HCC) with metastasis in 90% of mice. Myc overexpression alone causes multifocal liver cancer, however the addition of Twist1 results in lung, peritoneum and lymph node metastasis. Mice die by 25 months (as compared to 32 months with Myc alone).

Twist1-tetO7-luc transgenic mice carry a luciferase (luc) reporter and the mouse transcription factor Twist1, regulated by a tetracycline operator (tetO). They are an effective tool for generating inducible, tissue-specific, targeted mutants to study TWIST1 in inflammation, cancer development and metastasis. When bred with another transgenic mouse expressing tetracycline-controlled transactivator protein (rtTA or tTA), tissue-specific Twist1 expression in the resulting bitransgenic offspring can be regulated with the tetracycline analog, doxycycline.

A minimum of two mice must be ordered. Additional mice will be charged on a per-mouse basis. Click the “Details” button to learn more.

Our minimum guarantee for cryorecovery orders is 2 carriers. To ensure that your colony can be rapidly expanded upon arrival, additional carriers may be added to your order.
