Description

The FVB-Esrra^flox mice on the FVB strain background have loxP sites flanking exon 2 of the estrogen-related receptor, alpha (Esrra) gene. Exposure to Cre recombinase removes the floxed sequence, creating a null allele. These Esrra^f/f mice may be useful in studying oncology, immunology, and metabolism.

Detailed description

Esrra (estrogen related receptor, alpha) is a transcription factor involved in energy metabolism, with the highest expression observed in metabolically active tissues such as skeletal muscle and the heart. It is also overexpressed in various cancers, including breast, prostate, and ovarian cancers. The Esrra^flox allele has loxP sites flanking exon 2 of the Esrra gene.

Prior to introduction of Cre recombinase, mice heterozygous or homozygous for the Esrra^flox allele are viable and fertile with normal breeding, and have no gross physical or behavioral abnormalities. When Esrra^f/f are bred to mice that express Cre recombinase, the resulting offspring will have exon 2 deleted, creating a null allele in the cells/tissues as determined by the Cre promoter. For example, when bred to myeloid specific Cre-expressing mice (LysMcre; see Stock No. 004781), the resulting myeloid cell knock-out homozygotes were confirmed to have no ESRRA expression in bone marrow-derived macrophages (by western blot; see Racioppi et al. 2019).

Of note, the Jackson laboratory has the same Esrra^flox allele carrying mice on the C57BL/6J genetic background donated by the same investigator. B6-Esrra^flox mice (Stock No. 041114)

Development

The Esrra^flox allele was created in the laboratory of Dr. Donald P. McDonnell at Duke University. A targeting vector was constructed to insert a loxP site and an FRT-flanked neomycin selection cassette upstream of exon 2 of the estrogen related receptor, alpha (Esrra) gene, and another loxP site downstream of exon 2. The construct was electroporated into (129S6/SvEvTac x C57BL/6NCr)F1 -derived G4 ES cells and then microinjected into recipient blastocysts. The resulting mice were then bred to FLPe-expressing mice (Stock No. 003946) to remove the neo cassette and the line was subsequently backcrossed 6 generations to FVB/NJ mice (Stock No. 001800) prior to being donated to The Jackson Laboratory. Upon arrival, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize FVB/NJ oocytes.

Allele

Symbol: Esrra^tm1.1Dpm
Name: targeted mutation 1.1, Donald McDonnell
MGI accession ID: MGI:6393265
Generation method: Targeted
Attributes: Conditional ready (e.g. floxed)
Synonyms: ERRa^fl
Marker symbol: Esrra
Marker name: estrogen related receptor, alpha
Marker synonyms: Err1; ERRa; ERRalpha; Errra; ESRL1; Estrra; Nr3b1
Chromosome: 19
Strain of origin: (129S6/SvEvTac x C57BL/6NCrl)F1
Molecular note: The targeting vector was constructed to insert a loxP site and a FRT-flanked neomycin selection cassette upstream of exon 2 and another loxP site downstream of exon 2. Flp-mediated recombination removed the FRT-flanked neo cassette leaving exon 2 floxed.
