Expression of MYC in these tet-o-MYC transgenic mice is regulated by a tetracycline operator (tetO). MYC is a transcription factor involved in cellular proliferation and differentiation, and is often upregulated in tumors, leukemias and lymphomas. In founder line 36 the tet-o-MYC transgene inserted on the Y chromosome and is transmitted only through males. Hemizygous males are viable and fertile. When mated to a mutant strain expressing tetracycline-controlled transactivator protein (tTA or rtTA), expression of MYC protein may be regulated with tetracycline or its analog doxycycline (dox) in the double mutant offspring. When bred to mice carrying Tg(EmuSR-tTa)83Bop (as Stock No. <a href="https://www.jax.org/strain/034533">034533</a>) expressing tTA in T cells, bitransgenic offspring without doxycycline treatment die within 5 months of T cell lymphomas and acute myeloid leukemias. When doxycycline treatment is administered, most mice exhibit a sustained tumor regression. When bred to Tg(Cebpb-tTA)5Bjd/J mice (as Stock No. <a href="https://www.jax.org/strain/003563">003563</a>) expressing tTA in liver, withdrawal of dox from double transgenic mice results in invasive metastatic liver tumors, and all mice die within 2 weeks of onset. When doxycycline treatment is re-administered, rapid and sustained tumor regression is evident. Of note: in FVB/N-Tg(tetO-MYC)36aBop/J (Stock No. <a href="https://www.jax.org/strain/019376"">019376</a>) the tet-o-MYC transgene inserted on an autosomal chromosome. The transgene is transmitted through both males and females. In an attempt to offer alleles on well-characterized or multiple genetic backgrounds, alleles are frequently moved to a genetic background different from that on which an allele was first characterized. The phenotype summarized above is for the FVB/N background.
