Description

Fezf1 (Fez family zinc finger 1) knock-in/knock-out mice have an EGFP reporter and an hPLAP (human placental alkaline phosphatase) marker inserted into the coding region of the Fez family zinc finger 1 (Fezf1) gene. These mice may be useful for studying the regulation of olfactory system development and sensory neuron identity.

Detailed description

Fezf1 (Fez family zinc finger 1) encodes a zinc finger family transcription factor expressed early in mouse development and is involved in the specification and differentiation of several central neuronal cell types including the olfactory bulb, olfactory epithelium, and a subset of retinal starburst amacrine cells. A Fezf1 antisense long coding RNA, not studied in this line to date, has been implicated in progression of several cancers. Fezf1^GFP knockin/knockout mice have an EGFP reporter and an hPLAP (human placental alkaline phosphatase) inserted into the Fezf1 coding region. EGFP expression recapitulates Fezf1 expression across the olfactory epithelium and in the retina.  In homozygotes, axonal projections from olfactory sensory neurons fail to reach the olfactory bulb and instead form fasciculated bundles.  Heterozygous mice are viable and fertile, however, mice homozygous for the mutation die one to two days after birth.  These mice may be useful for studying the regulation of the central nervous system identity.

Development

The targeting construct contains an EGFP-IRES-hPLAP (human placental alkaline phosphatase) followed by a loxP-flanked PGK-neomycin cassette inserted into the coding region of the Fezf1 gene. The construct was electroporated into 129-derived E14a embryonic stem (ES) cells. Correctly targeted ES cells were injected into blastocysts. Heterozygous males were crossed to beta-actin-cre CD1 mice to remove the neomycin cassette. Mice were then intercrossed to generate homozygotes. The colony has been maintained on a mixed genetic background. Upon arrival, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize 129S1/SvImJ (Stock No. 002448) oocytes.
