Detailed description

The Th-TRAP EN63 bacTRAP transgenic strain, incorporating an EGFP-RPL10a ribosomal fusion protein targeted to the Th (tyrosine hydroxylase) gene, enables cell labeling, sorting, and affinity purification of cell-specific polysomal RNAs. The endogenous Th gene was not perturbed.

The Th-EGFP/Rpl10a transgene is expressed in a discrete subset of neurons, e.g. periglomerular neurons in the olfactory bulb (A16 cell group), scattered neurons in the caudate-putamen, neurons in the medial preoptic area (A15 cell group), zona incerta (A13 cell group), hypothalamus, ventral tegmental area (A10 cell group), substantia nigra pars compacta (A9 cell group) and locus coeruleus (A6 cell group).

In addition to Th, Ascl2 (achaete-scute family bHLH transcription factor 2) is present on BAC RP23-350E13. No information about possible expression of this gene is currently available.

Development

A bacterial artificial chromosome (BAC) that holds the tyrosine hydroxylase (Th) gene (RP23-350E13) was modified so that it expressed EGFP/Rpl10a under the regulation of the Th promoter. This modified BAC was microinjected into the pronucleus of a FVB/N zygote and randomly integrated into the genome of the mouse. Details of the process may be found in PMID 20885380  (Nat. Protoc. (2010) 5:1678-1696). Hemizygotes were bred with Swiss Webster mice by the donating laboratory.
