Bcl11b (B cell leukemia/lymphoma 11B; also called CTIP2, COUP-TF-interacting protein 2) is a zinc finger transcription factor that plays key roles in the development of the central nervous and immune systems. BCL11B is also highly expressed in the developing epidermis, where it plays a critical role in keratinocyte proliferation and late differentiation events, as well as in the development of the epidermal permeability barrier. Germline disruption of BCL11B results in an arrest of T-cell development at an immature, CD4-CD8- stage(s), with the complete absence of αβ T cells.Exon 4 of the mouse Bcl11b gene, encoding approximately 75% of the open reading frame and six zinc finger motifs, is flanked by loxP sites in these floxed CTIP2L2/Bcl11bL2 mutant mice. Cre recombinase-mediated excision of the floxed region results in a knock-out allele, as determined by immunoblots of skin.
