Description

VAMP3 knock-out mice lack exons 1-3 of the vesicle-associated membrane protein 3 (Vamp3) locus. These mice exhibit reduced systolic blood pressure and altered urinary sodium excretion. This strain may be useful in the study of renal function, neuroinflammation, and vesicular trafficking.

Detailed description

Vesicle-associated membrane protein 3 (Vamp3) encodes a v-SNARE protein involved in solute carrier family 2 (facilitated glucose transporter), member 4 (previously known as Glut4) trafficking. Mice homozygous for the Vamp3 deletion are viable and fertile with normal bodyweight and growth characteristics. Homozygous knock-outs also exhibit lower systolic blood pressure, associated with transiently increased urinary sodium excretion and reduced expression of NKCC2, which co-localizes with VAMP3, in the thick ascending limbs (TALs) of the kidneys. Western blotting further demonstrates a hypomorphic effect of heterozygous deletion on VAMP3 protein expression. This strain may be useful for the study of neuroinflammation and astrocytic activation, oncology, and renal function.

Development

The VAMP3 KO allele was created in the laboratory of Dr. Jeffrey Pessin at University of Iowa. The vector was designed to replace exons 1-3 with a neo selection cassette. The construct was electroporated into (129X1/SvJ x 129S1/Sv)F1-Kitl⁺-derived R1 embryonic stem (ES) cells. Correctly targeted ES cells were subsequently microinjected into recipient blastocysts. The chimeric mice produced were then bred to C57BL/6J mice and founders were verified by PCR. Littermates were bred together to generate homozygous Vamp3 knock-out mice. The Vamp3 knock-out strain was subsequently maintained on the B6;129 background prior to being donated to The Jackson Laboratory in 2019. Upon arrival, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes (Stock No. 000664).

Molecular note: 7.8 kb of sequence were replaced with a neomycin selection cassette. The deleted region included exon 2, which contains the guanosine of the translational start codon, and exon 3, whcih encodes the entire coiled-coil domain. Western blot analysis of homozygous mutant mice showed an absence of the normal protein product.
