Description

IL-1B^- homozygotes carry a knock-out allele of the mouse Il1b (interleukin 1 beta) gene, and may be useful in studying inflammation.

Detailed description

Il1b (interleukin 1 beta; IL-1β) is a potent pro-inflammatory cytokine that is critical for the in vivo survival, expansion, and effector function of IL-17-producing helper T (T(H)17) cells during autoimmune responses. This includes experimental autoimmune encephalomyelitis (EAE), a model of the inflammatory demyelination in multiple sclerosis.

The IL-1B^- knock-out allele was created through the introduction of a PGK-neomycin cassette to exon 4, resulting in an in-frame termination codon that precludes the translation of the receptor binding domain. Northern blot analysis detects no transcripts in LPS-stimulated peritoneal exudate cells (PECs) from homozygous mice. Pulse-chase labeling and immunoprecipitation studies of thioglycollate-elicited PECs demonstrate that no detectable protein is present in homozygotes. ELISA analysis of ATP-treated PECs also does not detect protein produced by this allele.

Mice deficient in IL-1β manifest impaired contact hypersensitivity to trinitrochlorobenzone.

IL-1β-deficient and wildtype mice do not differ significantly in the numbers of peripheral blood erythrocytes, lymphocytes, monocytes, neutrophils, or platelets. Similarly, the cell content and architecture of the spleen and thymus are grossly similar.

Development

A PGK-neomycin resistance cassette was inserted into exon 4 of the mouse Il1b gene via homologous recombination in 129S2/SvPas-derived D3 embryonic stem (ES) cells. This resulted in an in-frame termination codon upstream of the IL-1β converting enzyme (ICE) cleavage site encoded by exon 5, precluding the translation of the receptor-binding domain of the protein. This line was backcrossed to BALB/c by the developing laboratory for an unknown number of generations.

Molecular note: A PGK-neomycin resistance cassette was inserted into exon 4 that resulted in an in-frame termination codon upstream of the ICE cleavage site. The authors predict that the receptor-binding domain of the protein would not be translated. Northern blot analysis detected no transcripts in LPS-stimulated peritoneal exudate cells from homozygous mice. Pulse-chase labeling and immunoprecipitation studies of thioglycollate-elicited PEC demonstrated that no detectable protein was present in homozygous mice. ELIZA analysis of ATP-treated PEC also did not detect protein produced by this allele.
