The mouse Drd2 (dopamine receptor D2) promoter drives expression of codon-optimized Flp recombinase in these Drd2-Flpo BAC transgenic mice. No other genes are reported to be expressed from the BAC.The donating laboratory reports that this strain labels most D2 medium spiny neurons (MSNs), but not all. They capture between 30-40% of all dopamine- and cAMP-regulated phosphoprotein, Mr 32 kDa (DARPP32)+ MSNs, indicating 10-20% of D2 MSNs are unlabeled. D1 MSNs are not labeled, nor are axons in the pars reticulate of the substantia nigra (SNr). Specificity is reported to be imperfect, as the donating laboratory picks up some DARPP32-/Flp+ cells. That said, co-staining for interneuron markers fails to reveal interneurons co-labeled with immunoglobulins. They suggest there may just be too few cells to sample/characterize. One possibility, they suggest, is that the DARPP32 stain may have also have been imperfect, and that some of these cells are actually unstained MSNs. Their in situ studies, however, are unequivocal in showing some Flp+ neurons that co-label for parvalbumin (PV) or choline acetyltransferase (ChAT). Staining in dopaminergic neurons (DANs) is to be expected. The donating laboratory has observed heterogeneity in penetrance in the dorsal-ventral axis, though it is unclear if this is related to Flp reporter, or whether they really have less Flp made dorsally as opposed to ventrally. They confirmed strong expression using a viral vector that expresses Flp-dependent channelrhodopsin-2 (ChR2), demonstrating that adult mice, in addition to developing mice, express Flp recombinase.Transgenic mice have no overt phenotype. Hemizygotes are viable and
fertile. Homozygous viability/fertility was not assessed by the donating
laboratory (October, 2019).

These transgenic BAC mice express FlpO recombinase from the mouse Drd2 (dopamine receptor D2) promoter. Reporter strain crosses indicate that most, but not all D2 medium spiny neurons (MSNs) can be labeled. Specificity is described as "imperfect" by the donating laboratory, but overall expression patterns reproduce that of endogenous D2Rs.

Typically mice are recovered in 12-16 weeks. Contact Customer Service to place an order or for more information.
