Description

MARCKS^F mutant mice possess loxP sites flanking exon 2 of the Marcks (myristoylated alanine rich protein kinase C substrate) gene and may be useful in generating conditional mutations to study the regulation of cell permeability and secretion.

Detailed description

Marcks (myristoylated alanine rich protein kinase C substrate) encodes a protein that functions as a substrate for both conventional and atypical isoforms of protein kinase C and is involved in the regulation of cell permeability and secretion.  MARCKS is expressed throughout the brain and is localized to the plasma membrane. These conditional MARCKS^F mice possess loxP sites flanking exon 2 of the Marcks gene. Mice that are homozygous for this floxed allele are viable and fertile. When bred to mice that express tissue-specific Cre recombinase, resulting offspring will have the exon 2 deleted in Cre-expressing tissues.

For example, when bred to mice with Cre recombinase expression in the ependymal cells, offspring exhibit intracellular accumulation of mucins, elevated oxidative stress, lipid droplet build up, increased numbers of reactive astrocytes, microglia and macrophages in interstitial brain tissue.

Development

The targeting construct was designed to insert a loxP site upstream of exon 2 and loxP-flanked neomycin cassette downstream of exon 2. The construct was electroporated into C57BL/6-derived embryonic stem (ES) cells. Transient transfection with Cre recombinase excised the neomycin resistance cassette, leaving exon 2 floxed. Correctly targeted ES cells were injected into blastocysts. Chimeric males were crossed to C57BL/6NTac females. The colony was crossed to C57BL/6NTac for approximately one generation. Upon arrival, mice were bred to C57BL/6NJ for at least one generation to establish the colony.

Allele

Symbol: Marcks^tm2.1Pjb
Name: targeted mutation 2.1,Perry J Blackshear
MGI accession ID: MGI:5770609
Generation method: Targeted
Attributes: Conditional ready (e.g. floxed); No functional change
Synonyms: MARCKS^F
Marker symbol: Marcks
Marker name: myristoylated alanine rich protein kinase C substrate
Marker synonyms: 80K-L; KINC; MACS; PKCSL; PRKCSL
Chromosome: 10
Strain of origin: C57BL/6
Molecular note: A loxP site was inserted upstream of exon 2. A floxed neomycin resistance cassette was inserted downstream of exon 2. Cre-mediated recombination removed the selection cassette and left exon 2 floxed.
