Description

The Bhlhe40 conditional knockout (cko) strain may be useful in applications related to circadian rhythms, cellular growth, immune responses, and transcriptional regulation.

Detailed description

Bhlhe40 (basic helix-loop-helix family, member e40) is a modulator of Bdnf transcription and neuronal excitability. Knock-outs of the gene have been shown to raise hippocampal levels of Bdnf mRNA and increase status epilepticus susceptibility. They have also demonstrated delayed re-entrainment in circadian models of jet lag.

Exons 1-4 of the mouse Bhlhe40 (basic helix-loop-helix family, member e40; also called Sharp-2, Bhlhb2, and Dec1) gene are flanked by loxP sites in these Bhlhe40 cko mice mice. Cre recombinase-mediated excision of the floxed region results in a knock-out.

Development

A lox2272 site was introduced upstream of exon 1 and a lox2272-FRT-PGKneo-FRT cassette was placed in intron 4 via homologous recombination in 129S1/Sv-p⁺Tyr⁺Kitl⁺-derived CJ7 embryonic stem (ES) cells. Resultant chimeric males were crossed with C57BL/6 females. The neomycin cassette was excised through crosses with an Actb-Flp strain. This strain was backcrossed to C57BL/6 for an unknown number of generations.

Allele

Symbol: Bhlhe40^tm1Rhli
Name: targeted mutation 1, Robert H Lipsky
MGI accession ID: MGI:3775802
Generation method: Targeted
Attributes: Conditional ready (e.g. floxed); No functional change
Marker symbol: Bhlhe40
Marker name: basic helix-loop-helix family, member e40
Marker synonyms: Bhlhb2; C130042M06Rik; Clast5; CR8; cytokine response gene 8; Dec1; eip1 (E47 interaction protein 1); HLHB2; SHARP2; SHARP-2; Stra13; Stra14
Chromosome: 6
Strain of origin: 129S1/Sv-Oca2⁺ Tyr⁺ Kitl⁺
Molecular note: An frt-flanked neo cassette with a 5' loxP site was inserted downstream of exon 4 and an additional loxP site was inserted upstream of exon 1. Germ-line flp-mediate recombination was used to remove the neo cassette leaving exons 1 through 4 floxed.
