Description

Gal^cKO mice have loxP sites flanking exon 3 of the galanin and GMAP prepropeptide (Gal) gene. These mice are useful for studying the role of galanin in coping behaviors.

Detailed description

Gal^cKO mice have loxP sites flanking exon 3 of the galanin and GMAP prepropeptide (Gal) gene. GAL encodes a neuroendocrine peptide that is widely expressed in the central and peripheral nervous systems, with expression also seen the gastrointestinal tract, pancreas, adrenal gland and urogenital tract. It has been implicated in regulating a variety of physiological processes including feeding, energy homeostasis, osmotic regulation, nociception, seizures, stress responses, cognition, and mood. Mice that are homozygous for this floxed allele are viable and fertile. When bred to mice that express tissue-specific Cre recombinase, resulting offspring will have exon 3 deleted in the Cre-expressing tissues. Cre-mediated removal of the floxed sequence results in a frameshift with exon 2 spliced to exon 4, eliminating expression of both galanin and galanin message associated protein (GMAP) which is expressed from the same mRNA.

Development

The L1L2_Bact_P cassette was inserted at a position on Chromosome 19 downstream of exon 2 of the galanin and GMAP prepropeptide (Gal) gene. The cassette is composed of, from 5’ to 3’, an FRT site, a lacZ sequence, a loxP site, and neomycin sequence followed by a second FRT site and a second loxP site. A third loxP site was inserted downstream of exon 3. The mutation was generated in (129S6/SvEvTac x C57BL/6NCr)F1-derived G4 embryonic stem (ES) cells, and correctly targeted ES cells were injected into blastocysts from B6(Cg)-Tyr^c-2J/J mice. The resulting chimeric mice were bred to ACTB:FLPe mice (Stock No. 005703) to remove the lacZ and neomycin cassette. Resulting Gal^cKO mice were bred to C57BL/6J mice for at least 24 generations. Upon arrival at The Jackson Laboratory, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes (Stock No. 000664).

The cassette is composed of, from 5' to 3', an FRT site, a lacZ sequence, a loxP site, and neomycin sequence followed by a second FRT site and a second loxP site. A third loxP site was inserted downstream of exon 3. Flp-mediated recombination removed the neo cassette leaving exon 3 floxed.
