Description

PigU floxed mice have loxP sites flanking exon 2 of the phosphatidylinositol glycan anchor biosynthesis, class U (Pigu) gene. These mice are useful for studying GPI anchor biosynthesis.

Detailed description

PigU floxed mice have loxP sites flanking exon 2 of the phosphatidylinositol glycan anchor biosynthesis, class U (Pigu) gene. PigU encodes a component of the GPI transamidase complex and may be involved in the recognition of either the GPI attachment signal or the lipid portion of GPI. Mice that are homozygous for this allele are viable and fertile. When bred to mice that express tissue-specific Cre recombinase, resulting offspring will have exon 2 deleted in the Cre-expressing tissues.

Development

The L1L2_st1 cassette was inserted at a position on Chromosome 2 upstream of exon 2 of the phosphatidylinositol glycan anchor biosynthesis, class U (Pigu) gene. The cassette is composed of a frt-flanked lacZ sequence fused to a neomycin sequence followed by a loxP site. A second loxP site was inserted downstream of exon 3. The mutation was generated in C57BL/6N-derived JM8.N4 embryonic stem (ES) cells, and correctly targeted ES cells of clone HEPD0536_5_F10 were injected into blastocysts. The resulting chimeric mice were bred to C57BL/6NTac-Gt(ROSA)26Sor^{tm2(CAG-flpo,-EYFP)Ics}/Ics mice to remove the frt-flanked lacZ sequence and neomycin cassette. Resulting PigU floxed mice were bred to Lck-cre mice (Stock No. 003802) for at least 10 generations. The Lck-cre allele was bred out of the mice that arrived at The Jackson Laboratory. Upon arrival, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes (Stock No. 000664).
