Description

Green fluorescent protein of the inserted targeting vector is preferentially observed in some populations of intestinal tissue, but otherwise recapitulates normal expression patterns. This reporter strain may be used to detect and monitor cells committed to interleukin 10 (IL10) production.

Detailed description

This reporter strain may be used to detect and monitor cells committed to interleukin 10 production. Green fluorescent protein knocked into the gene faithfully recapitulates normal expression patterns but is preferentially observed in some populations of the intestinal tissue. Expression assayed by fluorescent-activated cell sorting (FACS) has been detected after activation in macrophages and dendritic cells as well as intraintestinal lymphocytes and lamina propria lymphocytes. Targeted allele expression is slightly reduced, as compared to wild-type. Although homozygotes have reduced IL10 expression, they are both viable and fertile.

Of note, a strain carrying the same allele on a C57BL/6J genetic background is available as Stock No. 008379.

In an attempt to offer alleles on well-characterized or multiple genetic backgrounds, alleles are frequently moved to a genetic background different from that on which an allele was first characterized. It should be noted that the phenotype of these BALB/cJ-congenic mice could vary from that originally described on a B6J genetic background. We may modify the strain description if necessary as published results become available.

Development

An IRES-GFP (internal ribosome entry site-green fluorescent protein) cassette followed by a loxP-flanked neomycin resistance cassette was inserted between the stop codon and polyadenylation signal of exon 5 of the gene. The targeting vector was electroporated into 129S6/SvEvTac-derived TC1 embryonic stem (ES) cells. Selected clones were transfected with cre recombinase to delete the neomycin resistance cassette. Targeted clones were injected into recipient blastocysts. Chimeric males showing germline transmission were bred to C57BL/6 mice. This line was backcrossed to C57BL/6Ncr for ten generations by the donating investigator before being sent to The Jackson Laboratory as Stock No. 008379. Some of these mice were obtained by Dr. David Rothstein (University of Pittsburgh) and were subsequently backcrossed to BALB/cJ mice (Stock No. 000651) for 10 generations to create this congenic strain.
