Description

Detailed description

Pex16^KO mice lack exon 3 of the peroxisomal biogenesis factor 16 (Pex16) gene. PEX16 encodes a peroxisomal membrane protein that plays a role in peroxisome organization and biogenesis. Peroxisomes are organelles involved in the catabolism of very long chain fatty acids through beta oxidation. Heterozygous mice are viable and fertile, homozygous mice are embryonic lethal.

Development

The L1L2_gt1 cassette was inserted at a position on Chromosome 2 upstream of exon 3 of the peroxisomal biogenesis factor 16 (Pex16) gene. The cassette is composed of a frt-flanked lacZ sequence fused to a neomycin sequence followed by a loxP site. A second loxP site was inserted downstream of exon 3. The mutation was generated in C57BL/6N derived JM8.N4 embryonic stem (ES) cells, and correctly targeted ES cells of clone HEPD0537_2_B03 were injected into blastocysts. The resulting chimeric mice were bred to C57BL/6NTac females. Cryopreserved sperm from this strain was sent to The Jackson Laboratory. To establish a live colony of Pex16^flox mice, lacking the frt-flanked lacZ sequence and neomycin cassette yet retaining floxed exon 3 (Stock No. 034155), an aliquot of frozen sperm was used to fertilize B6N.ROSA26::FLPe oocytes (Stock No. 016226). Subsequently, some resulting male offspring were bred to B6N.Sox2-Cre females (Stock No. 014094) to remove exon 3, thus creating this new Pex16^KO strain.
