Description

Pex16^flox mice have loxP sites flanking exon 3 of the peroxisomal biogenesis factor 16 (Pex16) gene. These mice are useful for studying peroxisome organization and biogenesis.

Detailed description

Pex16^flox mice have loxP sites flanking exon 3 of the peroxisomal biogenesis factor 16 (Pex16) gene. PEX16 encodes a peroxisomal membrane protein that plays a role in peroxisome organization and biogenesis. Peroxisomes are organelles involved in the catabolism of very long chain fatty acids through beta oxidation. Mice that are homozygous for this floxed allele are viable and fertile. When bred to mice that express tissue-specific Cre recombinase, resulting offspring will have exon 3 deleted in the Cre-expressing tissues.

Of note, a knock-out of this gene is available as Stock No. 034156.

Development

The L1L2_gt1 cassette was inserted at a position on Chromosome 2 upstream of exon 3 of the peroxisomal biogenesis factor 16 (Pex16) gene. The cassette is composed of a frt-flanked lacZ sequence fused to a neomycin sequence followed by a loxP site. A second loxP site was inserted downstream of exon 3. The mutation was generated in C57BL/6N derived JM8.N4 embryonic stem (ES) cells, and correctly targeted ES cells of clone HEPD0537_2_B03 were injected into blastocysts. The resulting chimeric mice were bred to C57BL/6NTac females. Cryopreserved sperm from this strain was sent to The Jackson Laboratory. To establish our live colony of Pex16^flox mice, lacking the frt-flanked lacZ sequence and neomycin cassette, an aliquot of frozen sperm was used to fertilize B6N.ROSA26::FLPe oocytes (Stock No. 016226).

Molecular note: The L1L2_gt1 cassette was inserted at a position on Chromosome 2 upstream of exon 3 of the gene. The cassette is composed of an FRT-flanked lacZ sequence fused to a neomycin sequence followed by a loxP site. A second loxP site was inserted downstream of exon 3. Flp-mediated recombination removed the FRT-flanked lacZ/neo cassette.
