H11-CAG-FRT-stop-FRT-EGFP mice express enhanced green fluorescent protein (EGFP) reporter from the Igs2 (intergenic site 2; H11) locus upon Flp recombinase-mediated excision of an FRT-flanked Stop cassette. H11 supports widespread, high-level EGFP gene expression from the CMV enhancer/chicken beta actin promoter.

Detailed description

H11-CAG-FRT-stop-FRT-EGFP mice express enhanced green fluorescent protein (EGFP) reporter from the Igs2 (intergenic site 2; also called H11) locus upon Flp recombinase-mediated excision of an FRT-flanked Stop cassette. H11 supports widespread, high-level EGFP gene expression from the CMV enhancer/chicken beta actin promoter. H11 knock-in mutations are associated with higher rates of mitotic (interchromosomal) recombination as compared to Gt(ROSA)26Sor.

When crossed with Sert-Flp mice (see Stock No. 034050), this strain has been useful in the labeling of serotonin-expressing cells.

Development

The H11-CAG-FRT-Stop-FRT-EGFP mouse line was generated through site-specific integrase-mediated transgenesis by injecting a CAG-FRT-Neo-Stop-FRT-EGFP-sv40-polyA transgene plasmid along with φC31 integrase RNA into the cytoplasm of FVB embryos carrying three tandem attP sites at the Igs2 (Hipp11, H11 P3) intergenic site (Igs2^tm3.1Luo, PMID 21464299). The primary Igs2^tm3.1Luo (H11 P3) mutation was created using (129X1/SvJ x 129S1/Sv)F1- Kitl⁺-derived R1 embryonic stem (ES) cells. This H11-CAG-FRT-Stop-FRT-EGFP strain was maintained on a mixed genetic background potentially incorporating FVB, CD1, 129S1/Sv, 129X1/SvJ, C57BL/6J, and DBA/2J genetic contributions by the donating laboratory.
