Description

Sert-Flp mice express FlpO recombinase from the mouse Slc6a4 (solute carrier family 6 (neurotransmitter transporter, serotonin), member 4; also known as Sert) promoter in cells that express serotonin.

Detailed description

The serotonin system innervates nearly every region of the brain, even though it only constitutes ~1/200,000 of all CNS neurons in humans. It is critically involved in a broad range of brain functions and is the most frequently targeted neural system pharmacologically for treating psychiatric disorders. The dorsal raphae (DR) and median raphae (MR) serotonin systems have been linked with the regulation of many mental states and processes, including anxiety, mood, impulsivity, aggression, learning, reward, social interaction, and hence remain the focus of intense research.

These Sert-Flp targeted mutant mice express codon-optimized FlpO recombinase from the mouse Slc6a4 (solute carrier family 6 (neurotransmitter transporter, serotonin), member 4; also known as Sert) promoter and enable the excision of FRT-flanked sequences of interest in serotonin-expressing cells. Slc6a4 expression is not disrupted.

Characterization of the Sert-Flp mice was carried out through crosses with H11-CAG-FRT-Stop-FRT-EGFP reporter mice (see Stock No. 034051).

An intersectional strategy to label the serotonin neurons co-expressing markers for glutamate or GABA transmission, can be accomplished by crossing these Sert-Flp mice with IS tdTomato reporter mice (see Stock No. 028582) and either Vglut3-Cre (Slc18a8-Cre; see Stock No. 018147) or Gad2-Cre

Development

An IRES-FlpO-loxP2272-ACE-Cre POII NeoR-loxp2272 cassette was inserted after the TAA stop codon of Slc6a4/Sert via homologous recombination in 129S6 x C57BL/6J-derived embryonic stem (ES) cells. The loxP2272-ACE-Cre POII NeoR-loxp2272 portion of the mutation is self-excised. Resultant chimeric males were bred with CD1 females. This line was backcrossed to C57BL/6 for 6 generations by the donating laboratory.
