Product name: B6 B-actinCre
Research areas: Research Tools

Description

These β-actin-cre transgenic mice express Cre recombinase directed by the human beta actin gene promoter. Cre recombinase activity is widespread in the embryo by the blastocyst stage of development.

Of note, this allele is also available on a congenic C57BL/6N genetic background as Stock No. 019099 and on a congenic FVB/N background as Stock No. 003376.

Detailed description

These β-actin-cre transgenic mice express Cre recombinase under the control of the human beta actin gene promoter. Cre recombinase activity is widespread in the embryo by the blastocyst stage of development. It is useful as a Cre-deleter mouse for virtually any genomic sequence flanked by loxP sites. The donating investigator has not observed any difference in phenotype between hemizygotes and homozygotes. Homozygotes are viable and fertile.

In an attempt to offer alleles on well-characterized or multiple genetic backgrounds, alleles are frequently moved to a genetic background different from that on which an allele was first characterized. This is the case for the strain above. It should be noted that the phenotype could vary from that originally described. We will modify the strain description if necessary as published results become available.

Of note, this allele is also available on a congenic C57BL/6N genetic background as Stock No. 019099 and on a congenic FVB/N background as Stock No. 003376.

Development

A transgenic insert designed by Drs. Mark Lewandoski and Gail R. Martin (University of California, San Francisco) containing Cre recombinase under the control of the human beta-actin promoter was injected into fertilized FVB/N mouse eggs. Founder line 2, containing a single copy of the transgene inserted into intron 4 of transmembrane protein 163 (Tmem163) gene, was subsequently established. The mice were maintained on the FVB/N background. Dr. Elizabeth M. Simpson (University of British Columbia) obtained the mice and backcrossed them to C57BL/6J mice (Stock No. 000664) for 15 generations. Cryopreserved sperm was sent to The Jackson Laboratory. Upon arrival, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes to establish our live colony.
