Description

These Bak flox mutant mice possess loxP sites flanking exons 3 and 4 of the Bak1 gene. This strain may be useful for generating conditional mutations in applications related to the regulation of the intrinsic pathway of apoptosis.

Detailed description

The targeted Bak1 gene encodes a proapoptotic Bcl-2 family member and regulates intrinsic pathway of apoptosis via mitochondrial membrane permeability. These mice possess loxP sites on either side of exons 3 and 4 of the targeted gene. Mice that are homozygous for this allele are viable and fertile. When these mutant mice are bred to mice that express Cre recombinase, resulting offspring will have exons 3 and 4 deleted in the cre-expressing tissues. Removal of the floxed sequence creates a null allele.

When bred to a strain with Cre recombinase expression in renal proximal tubule, this mutant mouse strain may be useful in studies of renal interstitial fibrosis.

 

Development

A targeting vector containing a FRT site flanked Neo selection cassette was utilized in the construction of this mutant. This selection cassette and a loxP site were inserted upstream of exon 3 of the targeted gene, and another loxP site was inserted upstream of exon 4. This construct was electroporated into unspecified C57BL/6-derived embryonic stem (ES) cells. Correctly targeted ES cells were injected into blastocysts. The resulting chimeric animals were tested for germline transmission. The mice were crossed to FLPe deleter mice (Stock No. 003800) to remove the selection cassette. The mice may have been crossed to other lines containing BALB/c and 129P2/OlaHsd genetic background contributions. Upon arrival, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes (Stock No. 000664).
