Suv39h1 (suppressor of variegation 3-9 1) and Suv39h2 (suppressor of variegation 3-9 2) encode histone methyltransferases involved in transcriptional gene silencing by trimethylation of lysine 9 of histone H3 (H3k9me). These methyltransferases and a third, Setdb1, are associated with H3k9me3-marked heterochromatin (compacted chromatin). CRISPR/Cas9 endonuclease mediated genome editing introduced loxP sites flanking exons 3-5 into Suv39h1 and a 4 base pair deletion in exon 2 of Suv39h2. Double mutant mice homozygous for Suv39h2KO and homozygous females/hemizygous males for Suv39h1fl are viable and fertile. When bred to mice that express tissue-specific Cre recombinase, resulting offspring will have the exons 3-5 of Suv39h1 deleted in cre-expressing tissues. These mice may be useful for studying H3k9me3-marked heterochromatin and lineage commitment and fidelity maintenance.
