Description

Ape1^LoxP mutant mice possess loxP sites flanking exons 2 and 3 of the Apex1 gene. This strain may be useful for generating conditional mutations in applications related to the repair of oxidative DNA damage.

Detailed description

The targeted Apex1 gene (also known as Ape1) encodes apurinic endonuclease, an oxidative DNA damage repair enzyme. Mutations in this gene have been associated with Pediatric T-Cell Leukemia and Galloway-Mowat Syndrome 3.
These Ape1^LoxP mice possess loxP sites flanking exons 2 and 3, which include the initiation ATG codon and second coding exon, of the targeted gene. Mice that are homozygous for this allele are viable and fertile.
When these mutant mice are bred to mice that express Cre recombinase, resulting offspring will have exons 2 and 3 deleted in the cre-expressing tissues. Removal of the floxed sequence creates a null allele.
For example, when bred to a strain with Cre recombinase expression in the central and peripheral nervous system (e.g., see Stock No. 003771), this mutant mouse strain may be useful in studies of the repair of oxidative DNA lesions during neurogenesis.

Development

A targeting vector containing an FRT site flanked PGK-Neo selection cassette was utilized in the construction of this mutant. This selection cassette and a loxP site were inserted upstream of exon 2 of the targeted gene, and another loxP site was inserted downstream of exon 3. This construct was electroporated into 129S1/Sv-Oca2⁺ Tyr⁺ Kitl⁺ derived W9.5 embryonic stem (ES) cells. Correctly targeted ES cells were injected into blastocysts. The resulting male chimeric animals were crossed C57BL/6 female mice to establish germline transmission. The mice were then bred to B6;SJL-Tg(ACTFLPe)9205Dym/J transgenic mice expressing FLP1 recombinase under the control of the human ACTB promoter (Stock No. 003800). Mice that retained the loxP site flanked exons 2 and 3 (which includes the initiation ATG codon and second coding exon) were then bred to C57BL/6 mice to remove the FLPe transgene. Upon arrival, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes (Stock No. 000664).

Allele

Symbol: Apex1^tm1.1Pmc
Name: targeted mutation 1.1, Peter J McKinnon
MGI accession ID: MGI:6361426
Generation method: Targeted
Attributes: Conditional ready (e.g. floxed)
Synonyms: Ape1^loxP
Marker symbol: Apex1
Marker name: apurinic/apyrimidinic endonuclease 1
Marker synonyms: APE; APE1; APEN; APEX; APX; HAP1; REF1; REF-1
Chromosome: 14
Strain of origin: 129S1/Sv-Oca2⁺ Tyr⁺ Kitl⁺
Molecular note: The targeting vector contains an FRT-flanked neomycin selection cassette and a loxP site inserted in intron 1 and a second loxP site inserted downstream of exon 3. Flp-mediated recombination removed the FRT-flanked neo cassette leaving exons 2 through 3 floxed.
