Description

Ager-H2b:Venus knock-in reporter mice are suitable for use in applications related to tracking lung alveolar cell proliferation and differentiation.

Detailed description

Ager (advanced glycosylation end product-specific receptor) encodes a cell surface multiligand receptor that mediates interactions of advanced glycosylation end products (AGE) as well as molecules involved in homeostasis, development, and inflammation. Ager-H2b:Venus knock-in reporter mice express a Venus variant yellow fluorescent protein from the endogenous Ager locus. Venus is detected in Type 1 alveolar (AT1) epithelial cells using antibodies to GFP. AT1 cells cover 95% of the internal surface of the lung and are involved in gas exchange. Mice homozygous for the mutation are viable and fertile with no overt phenotype. These mice may be useful for tracking lung alveolar cell proliferation and differentiation.

Development

The targeting construct contains an H2b:Venus fusion protein (coding sequence for the human HIST1H2BB gene [histone 1 H2bb] with a C-terminal yellow fluorescent protein sequence) followed by a polyA and a FRT-flanked neomycin cassette inserted into the start codon of the Ager (advanced glycosylation end product-specific receptor) gene. The construct was electroporated into (129S6/SvEvTac x C57BL/6NCrl)F1-derived G4 embryonic stem (ES) cells. Correctly targeted ES cells were injected into blastocysts. The resulting chimeric animals were crossed to B6.Cg-Gt(ROSA)26Sor^{tm14(CAG-tdTomato)Hze}/J (Ai14). Offspring were backcrossed to C57BL/6 mice for an unknown number of generations by the donating lab (see SNP note below) and the Ai14 allele was bred out. Upon arrival, mice were bred to C57BL/6J for at least one generation to establish the live colony.

A 48 SNP (single nucleotide polymorphism) panel analysis, with 43 markers covering all 19 chromosomes and the X chromosome, as well as 5 markers that distinguish between the C57BL/6J and C57BL/6N substrains, was performed on the rederived living colony at The Jackson Laboratory Repository. Markers on Chormosomes 13 and 14 were segregating with an unknown source. Also, 4 of the 5 markers that determine C57BL/6J from C57BL/6N were found to be segregating. These data suggest the mice sent to The Jackson Laboratory Repository were on a mixed genetic background.
