Description

Fgfbp1^gfp mice have a GFP sequence replacing the Fgfbp1 coding region. These mice are useful when studying cell proliferation, inflammation, and tumor growth.

Detailed description

Fgfbp1^gfp mice have a green fluorescent protein (GFP) sequence replacing the entire coding region of the fibroblast growth factor binding protein 1 (Fgfbp1) gene. FGFBP1 plays a role in wound healing, and cell proliferation, differentiation and migration by binding to fibroblast growth factors. It has also been shown to be involved in tumor growth as an angiogenic switch molecule, and expression of this gene has been associated with several types of cancer including pancreatic and colorectal adenocarcinoma. Homozygous and heterozygous mice are viable and fertile. Heterozygous Fgfbp1^gfp mice revealed rapid induction of gene expression during papilloma induction and during wound healing.

Development

The Fgfbp1^loxP-neo-gfp targeting vector was designed to insert a loxP site upstream of the entire fibroblast growth factor binding protein 1 (Fgfbp1) gene coding region. A second loxP site and a frt site, followed by neomycin selection cassette (neo), a third loxP site, and a green fluorescent protein (GFP) sequence followed by a second frt site, were inserted downstream of the Fgfbp1 gene. The construct was electroporated into (C57BL/6N;129/SvEv)F1-derived embryonic stem (ES) cells. Correctly targeted ES cells were injected into C57BL/6N blastocysts and resulting chimeric mice were maintained on a mixed genetic background. Cryopreserved sperm was sent to The Jackson Laboratory. Upon arrival, an aliquot of sperm was used to fertilize oocytes from Sox2-cre mice (Stock No. 008454). Resulting Fgfbp1^gfp offspring have only the GFP sequence replacing the Fgfbp1 coding region. These mice are maintained on a mixed genetic background.
