The C5aR2-tg contains a human CD2 antigen (CD2) minigene driving expression of the complement component 5a receptor 2 (C5ar2) gene in CD4+ T cells. Homozygotes are viable, fertile and normal in size. C5L2 is a G-protein coupled receptor 1 family member involved in the complement system of the immune response. Expressed on immune cells, C5aR2 modulates various inflammatory diseases in mice, including sepsis, asthma, cystic fibrosis, and chronic obstructive lung disease, and has been shown to facilitate regulatory T cell (TREG) generation in vitro and in vivo. In these mice, overexpression of C5aR2 in immune cells prolongs graft survival associated with an increase in TREG/effector T cell ratios without altering effector T cell proliferation or cytokine production.
