Description

23-Apr

Casd1 floxed mice have loxP sites flanking exon 2 of the CAS1 domain containing 1 (Casd1) gene. These mice are useful for studying the requirement of acetylation of sialic acids to cell-cell interactions and host-pathogen recognition.

Detailed description

Casd1 floxed mice have loxP sites flanking exon 2 of the CAS1 domain containing 1 (Casd1) gene. Casd1 encodes an O-acetyltransferase that catalyzes 9-O-acetylation of sialic acids, which are sugars at the reducing end of glycoproteins and glycolipids, and are involved in various processes such as cell-cell interactions and host-pathogen recognition. Mice that are homozygous for this Casd1 floxed allele are viable and fertile. When bred to mice that express tissue-specific Cre recombinase, resulting offspring will have exon 2 deleted in the Cre-expressing tissues.

Deletion of Casd1 in the germline shows a loss of 9-O-acetylation of sialic acids on multiple hematopoietic lineages, yet without obvious defects in hematopoiesis. Casd1-deficient erythrocytes lose reactivity to TER-119, a rat monoclonal antibody used to mark the murine erythroid lineage.

Development

The L1L2_Bact_P cassette was inserted at a position on Chromosome 6 upstream of exon 2 of the CAS1 domain containing 1 (Casd1) gene. The cassette is composed of a frt site followed by a lacZ sequence and a loxP site. This first loxP site is followed by neomycin under the control of the human beta-actin promoter, SV40 polyA, a second frt site and a second loxP site. A third loxP site was inserted downstream of exon 2. The construct, clone HEPD0638_5_H05, was introduced into C57BL/6N-A^tm1Brd derived JM8A3.N1 embryonic stem (ES) cells, and correctly targeted ES cells were injected into blastocysts. The resulting chimeric mice were bred to C57BL/6NTac mice and then subsequently to FLPo-10 mice (Stock No. 011065) to remove the frt-flanked lacZ sequence and neomycin cassette. Resulting Casd1 floxed mice were bred to C57BL/6NTac mice for at least two generations. Upon arrival at The Jackson Laboratory, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6NJ oocytes (Stock No. 005304).

Molecular note: The L1L2_Bact_P cassette was inserted at position 4607550 of Chromosome 6 upstream of exon 2 (Build GRCm39). The cassette is composed of an FRT site followed by lacZ sequence and a loxP site. This first loxP site is followed by a neomycin resistance gene under the control of the human beta-actin promoter, SV40 polyA, a second FRT site and a second loxP site. A third loxP site is inserted downstream of exon 2 at position 4608344. FLP-mediated recombination removed the FRT-flanked neo cassette leaving exon 2 floxed.
