Description

Dgat2 flox mice possess loxP sites flanking exons 3 and 4 of the Dgat2 gene. This strain may be useful for generating conditional mutations in applications related to triglyceride synthesis and Charcot-Marie-Tooth Disease.

Detailed description

The targeted Dgat2 gene encodes an essential acyltransferase that catalyzes the final step of triglyceride synthesis. Mutations in this gene have been associated with Autosomal Dominant Charcot-Marie-Tooth Disease Type 2 Due To Dgat2 Mutation. These mice possess loxP sites on either side of exons 3 and 4 of the targeted gene. Mice that are homozygous for this allele are viable and fertile. When these mutant mice are bred to mice that express Cre recombinase, resulting offspring will have exons 3 and 4 deleted in the cre-expressing tissues. Removal of the floxed sequence creates a null allele.

When bred to a strain with Cre recombinase expression in the epidermis, this mutant mouse strain may be useful in studies of the epidermal water-permeability barrier.

Development

The Donating Investigator obtained the HEPD0666_3_A05 Dgat2^{tm1a(EUCOMM)Hmgu} ES cell line from EUCOMM and injected the ES cells into blastocysts. The resulting chimeric animals were backcrossed to wildtype C57BL/6J mice, and then to "actinb-FLPe" mice carrying the Gt(ROSA)26Sor^tm1(FLP1)Dym allele (Stock No. 005703) to generate Dgat2 flox mice, which have loxP sites flanking exons 3 and 4. The mice were then backcrossed to C57BL/6J for six generations. Upon arrival, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes (Stock No. 000664).

Attributes: Conditional ready (e.g. floxed)
Synonyms: Dgat2^flox
Marker symbol: Dgat2
Marker name: diacylglycerol O-acyltransferase 2
Marker synonyms: 0610010B06Rik; ARAT; GS1999FULL; HMFN1045
Chromosome: 7
Strain of origin: C57BL/6N-A^tm1Brd
Molecular note: The L1L2_Bact_P cassette was inserted at position 98813559 of Chromosome 7 upstream of exon 3 (Build GRCm39). The cassette is composed of an FRT site followed by lacZ sequence and a loxP site. This first loxP site is followed by a neomycin resistance gene under the control of the human beta-actin promoter, SV40 polyA, a second FRT site and a second loxP site. A third loxP site is inserted downstream of exon 4 at position 98814834. Flp-mediated recombination removed the FRT-flanked neo cassette leaving exons 3-4 floxed.
