Description

Sirt5^floxed mice possess loxP sites flanking exon 4 of the sirtuin 5 (Sirt5) gene. These floxed mice are useful when studying metabolic homeostasis.

Detailed description

Sirt5^floxed mice possess loxP sites flanking exon 4 of the sirtuin 5 (Sirt5) gene. SIRT5 is a mitochondrial NAD⁺-dependent deacylase with desuccinylase and demalonylase activities that is involved in metabolic homeostasis. Mice that are homozygous for this floxed allele are viable and fertile. When bred to mice that express tissue-specific Cre recombinase, resulting offspring will have exon 4 deleted in the cre-expressing tissues.

For example, when bred to mice with cardiac Cre recombinase expression, resulting Sirt5^-/- offspring exhibit defective fatty acid metabolism, decreased ATP production, and hypertrophic cardiomyopathy.

Development

The Sirt5^floxed targeting vector was designed to insert a loxP site upstream of exon 4, and second loxP site, followed by a frt-flanked neomycin selection cassette, downstream of exon 4 of the sirtuin 5 (Sirt5) gene. The construct was electroporated into 129Sv-derived embryonic stem (ES) cells. Correctly targeted ES cells were injected into C57BL/6J blastocysts and resulting chimeric males were bred with ACTFLPe mice on a C57BL/6J background (Stock No. 005703) to remove the neo cassette. Progeny were crossed to remove the Flp-expressing transgene and mice were subsequently bred to C57BL/6J mice (Stock No. 000664) for at least five generations. Upon arrival at The Jackson Laboratory, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes.
