Description

GCN5^flox mice possess loxP sites flanking exons 5-6 of the K(lysine) acetyltransferase 2A (Kat2a) gene. These floxed mice are useful when studying the role of GCN5 as a transcriptional activator.

Detailed description

GCN5^flox mice possess loxP sites flanking exons 5-6 of the K(lysine) acetyltransferase 2A (Kat2a) gene. GCN5 is a histone acetyltransferase (HAT) that functions primarily as a transcriptional activator and as a repressor of NF-kappa-B. Mice that are homozygous for this floxed allele are viable and fertile. When bred to mice that express tissue-specific Cre recombinase, resulting offspring will have exons 5-6 deleted in the cre-expressing tissues.

Development

The GCN5^flox targeting vector was designed to insert a loxP site upstream of exon 5, and second loxP site, followed by a frt-flanked neomycin selection cassette, downstream of exon 6 of the K(lysine) acetyltransferase 2A (Kat2a) gene. The construct was electroporated into 129Sv-derived embryonic stem (ES) cells. Correctly targeted ES cells were injected into C57BL/6J blastocysts and resulting chimeric males were bred with ACTFLPe mice on a C57BL/6J background (Stock No. 005703) to remove the neo cassette. Progeny were crossed to remove the Flp-expressing transgene and mice were subsequently bred to C57BL/6J mice (Stock No. 000664) for at least 10 generations. Upon arrival at The Jackson Laboratory, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes.

Synonyms: GCN5^flox; Gcn5^L2
Marker symbol: Kat2a
Marker name: K(lysine) acetyltransferase 2A
Marker synonyms: 1110051E14Rik; GCN5; Gcn5l2; hGCN5; PCAF-b; PCAF-B/GCN5

Molecular note: The targeting vector was designed to insert a loxP site upstream of exon 5, and second loxP site, followed by an FRT-flanked neomycin selection cassette, downstream of exon 6. Flp-mediated recombination removed the FRT-flanked neo cassette leaving exons 5 and 6 floxed.
