Description

NCoR1^L2 mice possess loxP sites flanking exon 11 of the nuclear receptor co-repressor 1 (Ncor1) gene. These floxed mice are useful when studying the role of NCoR1 as a negative transcriptional regulator.

Detailed description

NCoR1^L2 mice possess loxP sites flanking exon 11 of the nuclear receptor co-repressor 1 (Ncor1) gene. NCoR1 is a transcriptional repressor involved in metabolism, cellular development, and cancer. Mice that are homozygous for this floxed allele are viable and fertile. When bred to mice that express tissue-specific Cre recombinase, resulting offspring will have exon 11 deleted in the cre-expressing tissues.

For example, when bred to Lck-Cre mice (Stock No. 003802), expressing cre recombinase in thymocytes, resulting knock-out offspring have reduced mature thymocyte numbers.

Development

The NCoR1^L2 targeting vector was designed to insert a loxP site upstream of exon 11, and loxP-flanked and frt-flanked neomycin selection cassette, downstream of exon 11 of the nuclear receptor co-repressor 1 (Ncor1) gene. The construct was electroporated into 129Sv-derived embryonic stem (ES) cells. Correctly targeted ES cells were injected into C57BL/6J blastocysts and resulting chimeric males were bred with ACTFLPe mice on a C57BL/6J background (Stock No. 005703) to remove the neo cassette. Progeny were crossed to remove the Flpe-expressing transgene and mice were subsequently bred to C57BL/6J mice for at least 15 generations. Upon arrival at The Jackson Laboratory, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes (Stock No. 000664).
