Description

These Myf5^MyoD knock-in mice express mouse Myod1 from the endogenous Myf5 locus. They are suitable for use in applications related to the study of axial skeleton development.

Detailed description

Myf5 encodes the earliest expressed transcription factor with a major role in skeletal muscle differentiation and myogenesis. Mutations in this gene have been associated with ophthalmoplegia, external, with rib and vertebral anomalies. Myod1 encodes a myogenic transcription factor that regulates muscle cell differentiation and is involved in muscle regeneration. These Myf5^MyoD knock-in mice express mouse Myod1 from the endogenous Myf5 locus. Homozygotes are viable and fertile. At E10.5, homozygous embryos exhibit a wildtype Pdgfa (platelet derived growth factor, alpha) gene expression pattern.

Development

A targeting vector designed by Dr. Philippe Soriano (Fred Hutchinson Cancer Research Center) containing a floxed PGKneo, bovine growth hormone polyadenylation sequence, and PGKDTA was used to insert 1.8 kb mouse Myod1 cDNA into the first exon of the Myf5 gene. The construct was electroporated into 129S4/SvJaeSor-derived AK7 embryonic stem (ES) cells. Correctly targeted ES cells were injected into blastocysts. The resulting chimeric animals were tested for germline transmission. The mice were crossed to R26Cre (general cre deleter) mice to remove the floxed selection cassette. The mice were then bred to remove the Cre recombinase allele, and backcrossed to C57BL/6 for 10 generations. Upon arrival, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes (Stock No. 000664).
