Description

Ncoa4^flox mutant mice possess loxP sites flanking exons 2 through 6. This strain may be useful for generating conditional mutations in applications related to iron homeostasis and autophagy.

Detailed description

The targeted Ncoa4 (nuclear receptor coactivator 4) gene, also known as RET-Activating Gene ELE1, encodes a coactivator of the androgen receptor, and is involved in iron homeostasis and autophagy. These mice possess loxP sites flanking exons 2 through 6 of the targeted gene. Mice that are homozygous for this allele are viable and fertile. When these mutant mice are bred to mice that express Cre recombinase, resulting offspring will have exons 2 through 6 deleted in the cre-expressing tissues. Removal of the floxed sequence creates a null allele.

Development

A targeting vector containing a FRT site flanked Neo selection cassette was utilized in the construction of this mutant. This selection cassette was inserted downstream of exon 6 of the targeted gene, and another loxP site was inserted upstream of exon 2. This construct was electroporated into unspecified embryonic stem (ES) cells. Correctly targeted ES cells were injected into blastocysts. The resulting chimeric animals were tested for germline transmission. The mice were crossed to FLP deleter mice (on the C57BL/6J genetic background). Mice that retained the loxP site flanked exons 2 through 6 were then bred to remove the FLP recombinase expression mutation. Heterozygotes were crossed to generate homozygotes. Upon arrival, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes (Stock No. 000664).

Allele

Symbol: Ncoa4^tm1.1Jdom
Name: targeted mutation 1.1, Joseph Mancias
MGI accession ID: MGI:6306108
Generation method: Targeted
Attributes: Conditional ready (e.g. floxed)
Synonyms: Ncoa^fl
Marker symbol: Ncoa4
Marker name: nuclear receptor coactivator 4
Marker synonyms: 1110034E15Rik; 4432406M01Rik; ARA70; ELE1; PTC3; RFG
Chromosome: 14
Strain of origin: Not Specified
Molecular note: The targeting vector contains an FRT-flanked Neo selection cassette inserted downstream of exon 6 and another loxP site inserted upstream of exon 2. Flp-mediated recombination removed the FRT-flanked neo cassette leaving exons 2 through 6 floxed.
